Chronic granulomatous disease

Overview

Chronic granulomatous disease (CGD) is a primary immunodeficiency disorder characterized by recurrent and life-threatening bacterial and fungal infections due to defective phagocyte nicotinamide adenine dinucleotide phosphate (NADPH) oxidase enzyme activity. 848

Diagnosis

  • Clinical Presentation: Recurrent bacterial and fungal infections, often involving the skin, lungs, lymph nodes, and gastrointestinal tract. 848
  • Laboratory Tests: Nitroblue tetrazolium (NBT) or dihydrorhodamine 123 (DHR) flow cytometry to assess NADPH oxidase activity. 848
  • Genetic Testing: Identification of mutations in CYBB (gp91phox), CYBA (p22phox), NCF4 (p40phox), or NCF1 (p67phox) genes. 848
