Lethal faciocardiomelic dysplasia

Last edited: 4/16/2026

Overview

Lethal faciocardiomelic dysplasia is a newly identified autosomal recessive disorder characterized by severe congenital anomalies including dwarfism, distinctive facial features, profound cardiac defects, and limb malformations, typically leading to neonatal death due to cardiac failure 1.

Diagnosis

  • Key Diagnostic Criteria: Polyhydramnios, low birth weight, epicanthal folds, abnormal ears, microretrognathia, microstomia, microglossia, glossoptosis, webbed neck, severe cardiac defects, radial and ulnar hypoplasia, radial deviation of hands, brachymetacarpalia, thumb hypoplasia, clinodactyly, hypoplasia of the 5th finger, simian creases, fibular and tibial hypoplasia, talipes varus with hypoplastic heels, wide gap between 1st and 2nd toes, delayed bone age 1.
  • Recommended Tests: Detailed physical examination, echocardiography for cardiac defects, skeletal imaging (X-ray) to assess bone age and limb anomalies 1.
  • Management

  • First-Line Treatments: Supportive care focusing on respiratory and cardiac support, nutritional management 1.
  • Adjunctive Treatments: No specific pharmacological treatments mentioned; interventions are primarily supportive and symptomatic 1.
  • Special Populations

  • Pediatrics: Neonates exhibit severe symptoms leading to early mortality, primarily due to cardiac failure 1.
  • Comorbidities: Severe cardiac defects are central to the prognosis, with no specific management strategies beyond supportive care noted 1.
  • Key Recommendations

  • Consider lethal faciocardiomelic dysplasia in neonates with consanguineous parents presenting with a constellation of facial dysmorphisms, severe cardiac anomalies, and limb malformations, indicative of an autosomal recessive inheritance pattern (Evidence: Expert opinion) 1.
  • Perform comprehensive imaging studies including echocardiography and skeletal X-rays to confirm the diagnosis and assess the extent of anomalies (Evidence: Expert opinion) 1.
  • Provide intensive supportive care focusing on respiratory and cardiac support due to the high risk of neonatal mortality from cardiac failure (Evidence: Expert opinion) 1.
